If i do 'File in Options' then i get a error if i open the webpage.
Then i can open a file in the webshop but this file is not to find...

In a product, options tab, add option and choose file as the type.

Whats the error, does lt happen when you open the page or when you choose file or when you add to cart.
What type of file did you try to upload. what version of CS are you on.

Try to upload image with the small size. Most possibly the maximum size of the files which can be uploaded is limited on your server.
Also you should make sure that the 'var/custom_files' folder exists and has 777 permissions.
