I would like to ship a product via USPS Priority to a certain set of zip codes and USPS Ground to others using realtime postal information. Can this be done some how by setting up locations?
I can’t think of any way to do this - there is no way to relate realtime shipping methods to locations except using the Localization feature which only operates a the country level. It would be nice if the Localization feature were made more granular - perhaps allowing use of existing locations instead of specifying country.

Might it be possible using a custom shipping method? Instead of realtime?
You could do what you want with a manual shipping method since it allows you to specify different (or none) shipping charges for each location.
